Clinical information
Agomelatin is an anti-depressive agent with melatonergic effects and is administered for the treatment of episodes of major depressions. It has a double action principle: It is a highly affined agonist regarding MT1- and MT2-melatonin receptors and a competitive antagonist of the 5HT2C-serotonin receptor, which enhances the release of the neurotransmitters noradrenalin and dopamine in certain areas of the brain. The therapeutic effect is anti-depressive, soporific and normalises the circadian rhythm. The undesirable side effects can be fatigue, nausea, swooning and undue sweating.
